γ-Glutamylcysteine synthetase (γ-GCS, EC 6.3.2.2) is the rate-limiting enzyme in glutathione (GSH) synthesis, catalyzing the synthesis of γ-glutamylcysteine (γ-GC) from glutamate and cysteine. Studies have shown that this enzyme plays a role in cellular oxidative stress. In the presence of ATP and magnesium ions, γ-GCS catalyzes the synthesis of γ-glutamylcysteine (γ-GC) from glutamate and cysteine, while ATP dephosphorylates to produce inorganic phosphate molecules. The activity of γ-GCS can be calculated by measuring the rate of increase of inorganic phosphate.
