Protein is the ultimate executor of life functions. It is also an essential and widely used detection object in biomedical-related fields such as clinical diagnosis, disease typing, and drug screening. Traditional ELISA technology has always been the primary method in protein detection. However, although ELISA can be applied to almost all protein detection, its sensitivity is poor, generally only reaching the level of pg/mL. This makes the detection of low-abundance proteins often tricky. In drug research, it is found that the accurate detection of these ultra-low abundance proteins is of great significance for predicting the occurrence of diseases. Therefore, a protein detection technology with high sensitivity is urgently needed.

Single Molecular Array (SiMoA) technology is a newly developed technology that can be thousands of times more sensitive than ELISA. It brings protein detection technology directly into the era of single-molecule and digital detection and detects ultra-low abundance proteins with the accuracy of fg. The biological principle of Simoa detection is still the classical immunoreaction-double-antibody sandwich method, but the Simoa technology coats about 250,000 capture antibodies on small 2.7 μm magnetic beads. During detection, biotin-labeled detection antibody and avidin-coupled enzyme and substrate were added. A single magnetic bead was blocked in 238,000 4.5 μm reaction wells (Wells) through a layer of oil for reaction.
