Urease (UE, EC 3.5.1.5) is a nickel-containing oligomeric enzyme that specifically catalyzes the hydrolysis of urea to release ammonia and carbon dioxide. Urease activity is positively correlated with organic matter content, total nitrogen, and available nitrogen content, reflecting the nitrogen status. This kit uses the indophenol blue colorimetric method to determine the NH3-N produced from the hydrolysis of urea by urease. In a strong alkaline medium, ammonia reacts with hypochlorite and phenol to form the water-soluble dye indophenol blue, the intensity of which is proportional to the NH3-N content in the solution. This substance has a maximum light absorption at 578nm, allowing for the determination of urease activity.
