Tyrosine ammonia-lyase (TAL, EC 4.3.1.23) is a key enzyme in an alternative pathway for the synthesis of coumarin in organisms. In organisms, there are two pathways for the synthesis of coumarin. One is the conversion of phenylalanine to cinnamic acid by phenylalanine ammonia-lyase, followed by the cytochrome P450 enzyme system to produce coumarin. Some phenylalanine ammonia-lyases are found to directly catalyze tyrosine to produce coumarin, providing an alternative pathway for coumarin synthesis. This assay kit measures the activity of TAL by catalyzing L-tyrosine to produce coumarin, which has a maximum absorption at 310nm. The TAL activity is calculated by measuring the rate of increase in absorbance.
Additional Materials and Equipments Required
Microplate reader, 96-well plate (UV plate), water bath, centrifuge, pipette, mortar, ice, and distilled water. Note: Regular microplate cannot transmit UV light; a UV plate is required for detection wavelengths less than 340nm.
