Triose phosphate isomerase (EC 5.3.1.1, TPI or TIM) in plant chloroplasts is an important enzyme involved in the Calvin cycle of photosynthesis. Dihydroxyacetone phosphate can quickly pass through the chloroplast envelope into the cytoplasm and is gradually converted into sucrose there. TPI converts dihydroxyacetone phosphate to glyceraldehyde-3-phosphate, and then reacts with the enzyme mixture, accompanied by the generation of NADH. By detecting the increase rate of NADH at 340 nm, the activity of TPI enzyme can be calculated.
