Trehalose-6-phosphate synthase (TPS, EC 2.4.1.15) is one of the key enzymes in trehalose synthesis, catalyzing the conversion of UDP-glucose and glucose-6-phosphate to trehalose-6-phosphate and UDP. UDP and phosphoenolpyruvate are sequentially acted upon by pyruvate kinase and lactate dehydrogenase, oxidizing NADH to NAD+. By detecting the decrease in NADH at 340nm, the enzyme activity of TPS can be calculated.
