Protease activity is typically measured using natural and synthetic substrates. Various proteases can hydrolyze the natural substrate casein, so the activity measured from casein is the total protease activity. This assay kit utilizes total proteases to catalyze the hydrolysis of azocasein, and the products generated have light absorption at 366nm. The total protease activity can be determined by measuring the change in absorbance.
