Total Ascorbic Acid (TAA) includes both reduced and dehydroascorbic acid. Reduced ascorbic acid is easily oxidized to dehydroascorbic acid, which reacts with 2,4-dinitrophenylhydrazine to form a red complex soluble in sulfuric acid. This red substance has a maximum absorption peak at 520nm, from which the total ascorbic acid (TAA) content is calculated.

Visible spectrophotometer, 1mL glass cuvette (light path 1cm), concentrated sulfuric acid, mortar and pestle, ice, low-temperature centrifuge, pipette.
