Thioredoxin reductase (TrxR, EC 1.6.4.5) is a NADPH-dependent selenoenzyme with an FAD domain, a member of the pyridine nucleotide-disulfide oxidoreductase family, and together with thioredoxin and NADPH, constitutes the thioredoxin system. TrxR has similar activity to GR, catalyzing the reduction of GSSG to GSH, and is one of the key enzymes in the glutathione redox cycle. Thioredoxin reductase (TrxR) catalyzes the reduction of DTNB by NADPH to produce TNB and NADP+, TNB has a characteristic absorption peak at 412nm, and the increase rate of TNB at 412nm can be used to calculate the TrxR activity.
