Superoxide dismutase (SOD) (EC 1.15.1.1) is widely present in animals, plants, microorganisms, and cultured cells. It has anti-aging properties, improves the body's resistance to various diseases, and enhances the body's adaptability to the external environment. It is an important antioxidant enzyme in organisms. There are multiple methods for SOD activity determination, among which the NBT (nitro blue tetrazolium) method produces a formazan dye with poor water solubility that can interact with reduced xanthine oxidase, not reaching a 100% inhibition rate, thus affecting the sensitivity and accuracy of the detection. This kit uses the WST-8 method, which is more stable and sensitive. WST-8 reacts with superoxide anions (O2.-) produced by xanthine oxidase (XO) to form a water-soluble formazan dye with maximum absorption at 450nm. SOD can clear O2.-, thus inhibiting the formation of formazan. The deeper the color of the reaction liquid, the lower the SOD activity, and vice versa.
