When organisms are subjected to external stress, superoxide anions and other reactive oxygen species are produced and accumulated in large quantities, which can serve as a signal of oxidative stress in organisms. Therefore, the production of superoxide anion radicals in organisms under stress conditions can indirectly reflect the extent of tissue cell damage and resistance strength. Superoxide anions (O2-) react with hydroxylamine to produce NO2-, which under the action of p-aminobenzenesulfonic acid and α-naphthylamine, forms a pink azo dye that has the maximum light absorption at 540nm. The content of O2- in the sample can be calculated based on the A540 value.
