Sucrose is an important photosynthetic product, a primary substance for transport within plants, and one of the temporary storage forms of dominant carbohydrates. Sucrose synthase (Sucrose Synthase, EC 2.4.1.13) is a bi-directional enzyme that can catalyze both sucrose synthesis and degradation and is one of the key enzymes in sucrose metabolism. Studying the activity of SS-II in the synthesis direction is significant for sucrose synthesis in plants. SS-II catalyzes the reaction between free fructose and the glucose donor UDPG to produce sucrose. The colored product formed by the reaction of sucrose with resorcinol has a characteristic absorption peak at 480nm, and the enzyme activity is proportional to the depth of the color.
Additional Materials and Equipments Required
Visible spectrophotometer, 1 mL glass cuvette (light path 1cm), water bath, desktop centrifuge, pipette, mortar and pestle.
