Carbohydrates are one of the important components of plant bodies and the main raw materials and storage substances for metabolism. Under the action of concentrated sulfuric acid, carbohydrates undergo dehydration to form furfural or hydroxymethyl furfural, which then condenses with anthrone to form derivatives of furfural, a blue-green substance, with maximum absorption at 620nm in the visible light region, and its light absorption value is directly proportional to the sugar content within a certain range. This method is used for the determination of soluble monosaccharides, oligosaccharides, and polysaccharides, with the advantages of high sensitivity, simplicity, and quickness, and is suitable for the determination of trace samples. The method can determine almost all types of sugars (including monosaccharides: pentoses, hexoses, sucrose, glycogen, polysaccharides, etc.), so the sugar content measured by this method is the total soluble sugar content in the solution.

Visible spectrophotometer, 1mL glass cuvette (light path 1cm), boiling water bath, pipette, ethanol, concentrated sulfuric acid (self-prepared), mortar and pestle, and distilled water.
