Soil sucrase, also known as soil invertase, is most active in acidic media, and the sucrase commonly measured in soil is also an acidic invertase. It plays an important role in increasing soluble nutrients in soil and is related to soil organic matter, nitrogen, phosphorus content, microbial activity, and soil respiration intensity. Generally, the higher the soil fertility, the stronger the sucrase activity, making this enzyme an indicator for evaluating soil ripening and fertility levels. This kit uses the DNS colorimetric method, where soil sucrase catalyzes the degradation of sucrose to produce reducing sugars, which further react with 3,5-dinitrosalicylic acid to form colored amino compounds, with characteristic light absorption at 540nm. The increase rate of 540nm light absorption within a certain range is proportional to soil sucrase activity.
Additional Materials and Equipments Required
Visible spectrophotometer (wavelength setting: 540nm), 1mL glass cuvettes (optical path 1cm), desktop centrifuge, water bath/ incubator, pipettes, toluene.
