Nitrate nitrogen refers to the nitrogen element contained in nitrates. Organic matter in the soil decomposes to form ammonium salts, which are oxidized to nitrate nitrogen. Nitrate nitrogen in the soil is one of the main forms of nitrogen absorbed by higher plants, and its content is directly related to crop yield and quality. Nitrate ions in the soil extract have significant light absorption at 220nm and no absorption peak at 275nm. By measuring the absorbance of the soil extract at 275nm and multiplying it by a correction factor f to eliminate the light absorption interference of organic matter at 220nm, the content of nitrate nitrogen in the soil can be obtained.

Microplate reader, 96-well UV plate (for measurements under UV wavelengths), distilled water, balance, room temperature centrifuge, measuring cylinder, shaking incubator.
