Proteases are widely present in soil and can hydrolyze various proteins and peptides into amino acids, thus the activity of soil proteases is extremely important for the transformation of nitrogen in soil. Soil neutral protease (S-NPT) hydrolyzes casein to produce tyrosine under neutral conditions; tyrosine reacts with Folin-Ciocalteu reagent to form a blue compound under alkaline conditions; this blue substance has a characteristic absorption peak at 680nm, and thus the S-NPT enzyme activity can be obtained. Since the substrate casein itself contains various amino acids, a control with casein must be set during the detection process to subtract the interfering background values and exclude false positives.
