Soil exo-β-1,4-glucanase, also known as soil cellobiohydrolase (CBH, EC 3.2.1.91), is one of the components of the soil cellulase system. This enzyme acts on β-1,4-glycosidic bonds, cutting off a cellobiose (reducing sugar) molecule each time. This kit uses the enzyme to catalyze the reaction of p-nitrophenyl-β-D-cellobioside to produce the yellow substance p-nitrophenol (PNP), which has a characteristic light absorption at 405nm, thereby obtaining the activity of soil exo-β-1,4-glucanase (S-CBH).

Microplate reader (wavelength set to 405nm), 96-well plate, water bath or constant temperature shaking incubator, pipettes, distilled water.
