Asparaginase (ASNase, EC 3.5.1.1) is a type of amide enzyme that catalyzes the hydrolysis of asparagine into aspartic acid and ammonia, playing an important regulatory role in nitrogen metabolism. Soil asparaginase (S-ASNase) catalyzes the hydrolysis of asparagine into aspartic acid and ammonia. Ammonia, in a strong alkaline environment, reacts with hypochlorite and phenol to form the water-soluble dye indigo blue, which has a stable solution color. It has a characteristic absorption peak at 630nm, and by detecting the rate of increase of ammonia, the enzyme activity can be calculated.

Visible spectrophotometer, 1mL glass cuvettes (optical path 1cm), desktop centrifuge, pipette, water bath or constant temperature incubator, toluene and distilled water.
