Nitrogen is one of the main nutritional elements that limit plant growth and development, and soil nitrogen is the primary source of plant nitrogen nutrition. Soil arginine deaminase is also closely related to nitrogen transformation in soil. This kit utilizes arginine deaminase to hydrolyze arginine to generate NH3-N, which reacts with hypochlorite and phenol in a strong alkaline medium to form the water-soluble dye indigo blue. By detecting the maximum light absorption peak of the colored substance at 630nm, the activity of soil arginine deaminase can be determined.
Additional Materials and Equipments Required
Visible spectrophotometer, 1mL glass cuvettes (optical path 1cm), centrifuge, water bath or temperature-controlled incubator, pipettes, distilled water.
