SIRPA; mSIRP-alpha1; sirp-alpha-1; myD-1 antigen; Bit; P84; SIRP; SHP-1; CD172a; Ptpns1; SHPS-1
Accession
Predicted N Terminal
Thr 32
Form
Lyophilized from sterile PBS, pH 7.4, 5 % trehalose and 5 % mannitol.
Bio-activity
Measured by its binding ability in a functional ELISA. Immobilized mouse SIRPA-His at 10 μg/ml (100 μl/well) can bind human CD47-Fc, The EC50 of human CD47-Fc is 0.05-0.13 μg/ml.
Molecular Mass
Recombinant Mouse SIRPA comprises 353 amino acids and has a calculated molecular mass of 39.4 kDa. As a result of glycosylation, the apparent molecular mass of rmSIRPA is approximately 50-70 kDa in SDS-PAGE under reducing conditions.
Endotoxin
< 1.0 EU per 1 microgram of protein (determined by LAL method).
Purity
> 95 % by SDS-PAGE.
Storage
In lyophilized state for 1 year (4°C); After reconstitution under sterile conditions for 3 months (-70°C). Avoid repeated freeze/thaw cycles.
Reconstitution
Reconstitute in sterile distilled water to a concentration of 0.1-1.0 mg/mL.

Background
Immunoglobulin-like cell surface receptor for CD47. Acts as docking protein and induces translocation of PTPN6, PTPN11 and other binding partners from the cytosol to the plasma membrane. Supports adhesion of cerebellar neurons, neurite outgrowth and glial cell attachment. May play a key role in intracellular signaling during synaptogenesis and in synaptic function. Involved in the negative regulation of receptor tyrosine kinase-coupled cellular responses induced by cell adhesion, growth factors or insulin. Mediates negative regulation of phagocytosis, mast cell activation and dendritic cell activation. CD47 binding prevents maturation of immature dendritic cells and inhibits cytokine production by mature dendritic cells (By similarity).
