Ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco, EC 4.1.1.39) is a key enzyme in plant photosynthesis, controlling the fixation of CO2 and the distribution of carbon into the Calvin cycle and photorespiratory cycle, with its activity directly affecting the rate of photosynthesis. The initial activity reflects the activation state of the enzyme in vivo, and the total activity is the sum of the activated enzyme and the potential activation of the enzyme activity. Under the catalysis of Rubisco, one molecule of ribulose-1,5-bisphosphate (RuBP) combines with one molecule of CO2 to produce two molecules of 3-phosphoglycerate (PGA). With the combined action of 3-phosphoglycerate kinase and glyceraldehyde-3-phosphate dehydrogenase, glyceraldehyde-3-phosphate is produced, and reduced coenzyme I (NADH) is oxidized. Therefore, by detecting the decrease rate of NADH at 340 nm, the enzyme activity of Rubisco can be determined to synchronize the oxidation of NADH and the fixation of CO2. This kit includes an ATP regeneration system.
