Recombinant Human CD3D&CD3E Protein(Flag & His tag) (HH0280CL)

Recombinant Human CD3D Protein (P04234) extracellular domain (Met 1-Ala 105) with a flag tag at the C-terminus, constructed the plasmid 1; Recombinant Human CD3E Protein (P07766) extracellular domain (Met 1-Asp 126) with a polyhistidine tag at the C-terminus, constructed the plasmid 2. The two plasmids were co-expressed and the Human CD3D&CD3E heterodimer was purified.

CD3D; CD3 delta; T3D; CD3-DELTA; CD3E; T3E
Accession
Predicted N Terminal
Phe 22 & Asp 23
Form
Lyophilized from sterile PBS, pH 7.4, 5 % trehalose and 5 % mannitol.
Bio-activity
Measured by its binding ability in a functional ELISA. Immobilized human CD3D & CD3E Heterodimer Protein at 10 μg/ml (100 μl/well) can bind OKT3-mIgG2a. The EC50 of OKT3-mIgG2a is 7.0-18.0 ng/ml.
Molecular Mass
Recombinant heterodimer of Human CD3D&CD3E comprises 272 (126+146) amino acids and has a calculated molecular mass of 31.2 (14.4+16.8)KDa. As a result of glycosylation, the apparent molecular mass of Human CD3D&CD3E heterodimer is approximately 20, 22 and 26 KDa in SDS-PAGE under reducing conditions.
Endotoxin
< 1.0 EU per 1 microgram of protein (determined by LAL method).
Purity
(43.8+13.1+41.9) % by SDS-PAGE.
Storage
In lyophilized state for 1 year (4°C); After reconstitution under sterile conditions for 3 months (-70°C). Avoid repeated freeze/thaw cycles.
Reconstitution
Reconstitute in sterile distilled water to a concentration of 0.1-1.0 mg/mL.

Background
T-cell surface glycoprotein CD3 delta & CD3 epsilon chain, also known as CD3D & CD3E or CD3D&CD3E respectively, are single-pass type I membrane proteins. CD3D, together with CD3- epsilon(CD3E) , CD3-gamma and CD3-zeta, and the T-cell receptor alpha/beta and gamma/delta heterodimers, forms the T cell receptor-CD3 complex. T cell receptor-CD3 complex plays an important role in coupling antigen recognition to several intracellular signal-transduction pathways.
