Reducing sugars are widely present in animals, plants, microorganisms, and cultured cells. Reducing sugars in plants mainly include glucose, fructose, and maltose, which are the most common monosaccharides and disaccharides. Under alkaline conditions, the DNS reagent reacts with reducing sugars upon heating to form a brown-red amine compound. Scanning wavelengths from 480nm to 540nm reveals a characteristic absorption peak at 500nm. Within a certain concentration range, the content of reducing sugars is linearly related to the absorbance at 500nm. Based on the standard curve, the amount of reducing sugars in the sample can be determined.
Additional Materials and Equipments Required
Visible spectrophotometer, 1mL glass cuvette (light path 1cm), water bath, pipette, mortar and pestle, ethanol.
