Reduced glutathione (GSH) is the most abundant antioxidant thiol compound in cells and plays an important role in antioxidation, protein thiol protection, and amino acid transmembrane transport. The ratio of reduced to oxidized form (GSH/GSSG) is a major dynamic indicator of cellular redox status. Therefore, measuring the content of GSH and GSSG in cells, as well as the GSH/GSSG ratio, can well reflect the cellular redox state. Reduced glutathione (GSH) reacts with DTNB to form a complex with a characteristic absorption peak at 412nm; its absorbance is proportional to the GSH content.
