Pyruvate dehydrogenase (PDH, EC 1.2.4.1) is widely present in animals, plants, microorganisms, and cultured cells. It is the rate-limiting enzyme of the pyruvate dehydrogenase complex (PDHC) that catalyzes the oxidative decarboxylation of pyruvate, connecting glycolysis and the citric acid cycle. Pyruvate dehydrogenase (PDH) catalyzes the substrate sodium pyruvate to generate hydroxyethyl-TPP, which reduces methylene blue (MTT) to a blue product in the presence of the electron acceptor (PMS). The enzyme activity of PDH can be determined by measuring the increase rate of the blue product at 566nm.
