Pyruvate carboxylase (PC, EC 6.4.1.1) is widely present in animals, molds, and yeasts, but is not found in plants and most bacteria, mainly distributed in the mitochondria. Pyruvate carboxylase catalyzes the conversion of pyruvate to oxaloacetate and is a key enzyme in the TCA cycle for the replenishment of oxaloacetate, as well as the first rate-limiting enzyme in gluconeogenesis. Pyruvate carboxylase (PC) catalyzes the conversion of pyruvate, ATP, CO2, and water to oxaloacetate, ADP, and Pi. Malate dehydrogenase further catalyzes the conversion of oxaloacetate and NADH to malate and NAD+. The oxidation rate of NADH at 340nm can be measured to reflect the activity of PC.

UV-Vis spectrophotometer, 1mL quartz cuvette (light path 1cm), desktop centrifuge, pipette, mortar and pestle, ice, and distilled water.
