Pullulanase is a hydrolase widely present in microorganisms, animals, and plants, which can specifically break down the α-1,6-glucosidic bonds at the branching points of starch, glycogen, and their derivatives. Initially used for theoretical research on starch structure, by the 1970s, the application of pullulanase had expanded to various starch processing fields such as starch syrup, beer, and alcohol production, gradually moving from the laboratory stage to an industrial scale. Pullulanase catalyzes the decomposition of pullulan to produce reducing sugars, which further react with 3,5-dinitrosalicylic acid to form a brown-red amine compound with characteristic light absorption at 540nm. Within a certain range, the light absorption value at 540nm is proportional to the amount of reducing sugars generated. The activity of pullulanase is calculated by the rate of increase in light absorption.
Additional Materials and Equipments Required
Visible spectrophotometer, 1mL glass cuvette (light path 1cm), low-temperature centrifuge, water bath, pipette, mortar and pestle, ice, and distilled water.
