The ammonium nitrogen content in plant tissues can reflect the degree of plant stress. α-Amino acids, when heated with a solution of indole-3-ketone, undergo oxidative deamination to form the corresponding α-keto acids, which further decarboxylate to form aldehydes, while the indole-3-ketone is reduced. In a weak acid environment, the reduced indole-3-ketone reacts with ammonia and another molecule of indole-3-ketone to form a blue-purple complex, which has a characteristic absorption peak at 570nm.
