Phospholipase D (EC 3.1.4.4), also known as phosphatidylcholine hydrolase, is a collective term for enzymes that catalyze the hydrolysis and base exchange reactions of phosphodiester bonds, widely present in various organisms such as higher plants, animals, and bacteria, and participates in cellular lipid metabolism, signal transduction, biomembrane formation, and stress resistance. Phospholipase D catalyzes the hydrolysis of the substrate O-(4-nitrophenyl) choline (NPPC), and under the action of added acid phosphatase, produces p-nitrophenol (PNP), which has the maximum absorption peak at 405nm. By detecting the increase rate of PNP at 405nm, the activity of PLC can be determined.
