Monodehydroascorbate reductase (MDHAR, EC 1.6.5.4) is one of the key enzymes for the regeneration of ascorbate (AsA), catalyzing the reduction of MDHA back to AsA, which plays an important role in maintaining the antioxidant properties of ascorbate. MDHAR catalyzes the reduction of MDHA by NADH to produce AsA and NAD+. The activity of MDHAR is calculated by measuring the decrease in absorbance of NADH at 340 nm.
