Mitochondrial respiratory chain Complex V, commonly known as ATP synthase, F-type ATPase, and F1F0ATPase, is the ultimate reaction of mitochondrial oxidative phosphorylation. The main function of Complex V is to produce the energy ATP required by most cells. Abnormalities in this enzyme in animals can lead to heart and neurological diseases. Mitochondrial respiratory chain Complex V (F1F0ATPase) catalyzes the reaction of ADP and Pi to form ATP, accompanied by the reduction of NADP+ to NADPH through the successive action of hexokinase and glucose-6-phosphate dehydrogenase. The enzyme activity of mitochondrial Complex V can be determined by measuring the increase rate of NADPH at 340nm.

UV spectrophotometer, 1mL quartz cuvette (optical path 1cm), pipette, low-temperature desktop centrifuge, mortar and pestle, ice, and distilled water.
