Isocitrate is an isomer of citrate and, although present in small amounts, is widely found in the biological world. It is particularly abundant in the leaves of plants from the genus Bryophyllum or in raspberries and is also a component of the citric acid cycle. This assay kit provides a specific enzymatic method for detecting isocitrate content, using isocitrate dehydrogenase to convert isocitrate and NADP+ into α-ketoglutarate, carbon dioxide, and NADPH. The increase in absorbance at 340nm due to NADPH is measured to calculate the isocitrate content in the sample.
Additional Materials and Equipments Required
UV spectrophotometer, 1mL quartz cuvette (optical path 1cm), low-temperature centrifuge, pipette, mortar and pestle, and distilled water.
