Indole-3-acetic acid oxidase (IAAO) is an enzyme that plays an important role in regulating the level of IAA in plants. Its activity is crucial for adjusting the IAA level, which in turn affects the growth rate of the plant by oxidizing and inactivating IAA. The activity of IAAO can be represented by the rate at which it destroys IAA. In the reaction system, a quantified amount of IAA is converted into indole aldehyde under the action of IAAO, reducing the IAA content. The remaining IAA forms a red complex with FeCl3 in the presence of inorganic acid, which can be measured at 530 nm using colorimetry. The difference in IAA content between the blank and the enzyme solution can be used to calculate the activity of IAAO.
