Hemicellulose is a polysaccharide mixture that is closely combined with cellulose in the plant cell wall and is a major component of the primary cell wall. It is widely present in plants and is a new type of utilizable energy source. This kit, slightly improved based on the method of the US NREL laboratory, is used to detect the content of hemicellulose. Under acidic conditions and heating, hemicellulose is hydrolyzed to produce xylose. The content of xylose produced is detected by colorimetry, and the content of hemicellulose is calculated.

Microplate reader, 96-well plate, water bath, pipettes, 80% ethanol, acetone, concentrated sulfuric acid, mortar, and distilled water.
