Glycerol kinase (GK, EC 2.7.1.30) is the rate-limiting enzyme in glycerol metabolism, catalyzing the phosphorylation of glycerol to produce 3-phosphoglycerate, which is then converted to dihydroxyacetone phosphate by 3-phosphoglycerate dehydrogenase and enters the glycolytic pathway for oxidative breakdown and energy release. The deficiency of this enzyme directly leads to the cell's inability to utilize glycerol. This kit uses glycerol kinase to catalyze the Mg-ATP-dependent phosphorylation of glycerol to 3-phosphoglycerate, which is then converted to hydrogen peroxide by glycerol phosphate oxidase. Hydrogen peroxide reacts with the color reagent to have the maximum absorption peak at 510nm. By measuring the increase rate at 510nm, the enzyme activity of glycerol kinase can be determined.
