Glutathione reductase (GR, EC 1.6.4.2) is a flavoprotein oxidoreductase found in both animals and plants, catalyzing the reduction of oxidized glutathione (GSSG) to reduced glutathione (GSH). The higher the ratio of GSH/GSSG, the more reactive oxygen species produced during oxidative stress can be scavenged. Therefore, the activity of GR is an important indicator of the ability to cope with oxidative stress. This assay kit uses the Ellman method, where DTNB reacts with GSH produced by the reduction of GSSG by GR, generating a yellow product (TNB). This product has the maximum absorption at 412nm. The amount of TNB generated is linearly positively correlated with GR activity, and the activity level of glutathione reductase (GR) can be calculated by measuring the absorbance at 412nm. This method is more sensitive, stable, and operable than traditional test methods when measured in visible light. Additionally, the sample pretreatment is improved by adding corresponding reagents to eliminate the GSH content of the sample itself to ensure the accuracy of the test results.
