Glutathione peroxidase (GPx, EC 1.11.1.9) represents a family of enzymes with peroxidase activity, playing an important role in protecting organisms from oxidative damage. Traditional detection with hydrogen peroxide as the substrate can be interfered with by catalase, which decomposes hydrogen peroxide. The organic peroxide reagent (Cum-OOH) provided in this assay kit is not decomposed by catalase, thus specifically detecting the activity of glutathione peroxidase. This assay kit can quantitatively detect total glutathione peroxidase. GSH and DTNB react to produce a yellow substance, which has the maximum absorption peak at 412nm, while GSH-Px catalyzes the oxidation of GSH by the organic peroxide reagent (Cum-OOH), reducing the amount of GSH. The more GSH is reduced, the lighter the yellow of the reaction mixture, the higher the GSH-Px activity; conversely, the deeper the yellow, the lower the GSH-Px activity.
