Product description
Glutamine synthetase (GS, EC 6.3.1.2) is primarily found in plants and is one of the key enzymes in ammonia assimilation in living organisms. Inorganic nitrogen absorbed by plants, after being reduced to NH4+ by nitrate reductase (NR) and nitrite reductase (NIR), can only be assimilated and utilized through the GS/GOGAT pathway involving glutamine synthetase (GS). GS, in the presence of ATP and Mg2+, catalyzes the synthesis of glutamine from ammonium ions and glutamate; glutamine is further converted to γ-glutamyl hydroxamate, which forms a complex with a maximum absorption peak at 540nm under acidic conditions, allowing for the determination of GS enzyme activity.
Additional Materials and Equipments Required
Visible spectrophotometer, 1mL glass cuvettes (optical path 1cm), water bath, desktop centrifuge, pipettes, mortar and pestle, ice, and distilled water.
