Fumarase, also known as fumarate hydratase (EC 4.2.1.2), is one of the key enzymes in the citric acid cycle, found in mitochondria, and the cytosolic fumarase is closely related to the metabolism of amino acids and fumarates. In humans, the deficiency of this enzyme can lead to severe health problems, such as fetal brain malformations and hypotonia. Fumarase catalyzes the conversion of fumarate to L-malate, which is then reduced to NADH by malate dehydrogenase with NAD+, and the activity of fumarase can be determined by measuring the increase in NADH at 340nm.

UV-Vis spectrophotometer, 1mL quartz cuvette (light path 1cm), pipette, low-temperature centrifuge, mortar and pestle.
