The actions of fructan hydrolase (Levanase, EC 3.2.1.65) include the following points: hydrolyzing fructan to ensure energy supply, rapidly increasing osmotic pressure, and increasing the concentration of oligofructose under pressure, thereby maintaining matrix stability to achieve cold resistance. Fructan hydrolase hydrolyzes the substrate fructan to produce reducing sugars. The amount of reducing sugars is determined using the 3,5-dinitrosalicylic acid method, and the absorbance value is read at 540nm to determine the activity of fructan hydrolase.

Visible spectrophotometer, 1mL glass cuvette (light path 1cm), water bath, centrifuge, pipette, mortar and pestle, ice, and distilled water.
