Endo-β-1,4-glucanase (EC3.2.1.4) is found in bacteria, fungi, and animals and is one of the components of the cellulase system. This type of enzyme randomly hydrolyzes β-1,4-glycosidic bonds, shortening the amorphous long-chain cellulose molecules and breaking them down into various types of reducing sugars such as glucose, cellobiose, and cellotriose. Under alkaline conditions, the produced reducing sugars react with 3,5-dinitrosalicylic acid to form a brown-red substance with a maximum absorption peak at 540nm, from which the activity of endo-β-1,4-glucanase can be determined.
