Double-Luciferase Reporter Assay Kit (DEKIT011)

The Double-Luciferase Reporter Assay Kit is a tool designed for the simultaneous measurement of both luciferase activities within cells. The Firefly and Renilla luciferases are enzymes known for their bioluminescent properties. Firefly luciferase facilitates the conversion of luciferin into oxyluciferin, emitting light in the process. Similarly, Renilla luciferase catalyzes the oxidation of coelenterazine to coelenteramide, also resulting in bioluminescence. Initially, the activity of the firefly luciferase reporter gene is assessed in the presence of a fluorescein substrate. Subsequently, the fluorescence is quenched, and the coelenterazine substrate is introduced to trigger Renilla luciferase activity. This dual reporter system enhances experimental precision by enabling the detection of two distinct reporter genes concurrently. The kit boasts several advantages, including swift analysis, heightened sensitivity, a broad dynamic range, and minimal interference from endogenous cellular factors. This approach is particularly useful for comparative studies where two different gene expressions or regulatory elements are being evaluated in the same sample. The kit is suitable for samples from animals, plants, and protoplasts.
