Creatine kinase (CK, EC 2.7.3.2) is primarily found in the heart, muscle, and brain tissues, and catalyzes the reversible phosphotransfer reaction between creatine and ATP, playing an important role in energy metabolism, muscle contraction, and ATP regeneration. CK catalyzes the reaction of phosphocreatine and ADP to form creatine and ATP, followed by the sequential action of added hexokinase and glucose-6-phosphate dehydrogenase complex, catalyzing ATP hydrolysis and the generation of NADPH, detected by the increase in absorbance at 340nm to determine the enzyme activity.
