Pectin is a major component of the primary cell wall and middle lamella, mainly in the form of protopectin, pectic acid methyl ester, and pectic acid, widely distributed in plant fruits, roots, and leaves. Pectin is cross-linked with other substances through Ca2+, Mg2+, hydrogen bonds, glycosidic bonds, and ester bonds. Specific extraction methods can be used to extract ionically bound pectin (ISP) and covalently bound pectin (CSP). This kit specifically extracts covalently bound pectin (CSP) with a chelating agent's alkaline solution and uses the sulfuric acid-carbazole colorimetric method to determine the pectin content. Pectin is hydrolyzed to galacturonic acid, which undergoes a condensation reaction with carbazole in sulfuric acid solution to form a purple-red substance, with a maximum absorption peak at 530nm, the color depth of which is proportional to the pectin content, thus obtaining the content of covalently bound pectin.
