Chymotrypsin (EC 3.4.21.1), also known as trypsin, is a proteolytic enzyme secreted by the pancreas, acting as an endopeptidase that specifically hydrolyzes the carboxyl-terminal peptide bonds of tyrosine and phenylalanine in protein chains. Clinically, chymotrypsin is used for liquefaction of sputum, effective for both purulent and non-purulent sputum, and also for wound healing after trauma or surgery. This assay kit uses chymotrypsin to catalyze the hydrolysis of N-succinyl-Ala-Ala-Ala-Phe-p-nitroanilide (SAAPFpNA) to produce p-nitroaniline, which has a maximum absorption peak at 405nm. The enzyme activity of chymotrypsin can be determined by measuring the rate of increase in absorbance.
