β-1,3-Glucanase (β-1,3-GA, EC 3.2.1.39) is primarily found in plants, catalyzing the hydrolysis of β-1,3-glucosidic bonds, thereby disrupting fungal cell walls, especially in synergy with chitinases, which can significantly inhibit fungal growth. In plants under disease or other stress conditions, the synthesis of β-1,3-GA can be induced to enhance the plant's resistance to adverse external stimuli. Therefore, the activity assay of β-1,3-GA is widely used in plant pathology and stress physiology research. β-1,3-GA hydrolyzes the β-1,3-glucosidic bonds of laminarin, producing reducing ends. The amount of reducing sugars is determined using 3,5-dinitrosalicylic acid, and the absorbance at 540nm is read to determine the activity of β-1,3-glucanase.
