Phosphoenolpyruvate carboxykinase (PEPCK) belongs to the lyase family and is divided into two types: one is ATP-dependent, namely ATP-PEPCK (EC 4.1.1.49), primarily found in flowering plants, algae, and some fungi and bacteria. The other is GTP-dependent, namely GTP-PEPCK (EC 4.1.1.32), mainly found in mammals, birds, fish, insects, mollusks, flatworms, nematodes, euglenoids, and some fungi and bacteria. This assay kit measures the ATP-dependent PEPCK, which catalyzes the conversion of oxaloacetate and ATP to phosphoenolpyruvate and CO2. Subsequently, in the presence of pyruvate kinase and lactate dehydrogenase, NADH is oxidized to NAD+. By measuring the decrease in NADH at 340nm, the activity of PEPCK can be reflected.

UV-Vis spectrophotometer, 1mL quartz cuvette (light path 1cm), desktop centrifuge, pipette, mortar and pestle, ice, and distilled water.
