ATP-Citrate Lyase (ACL, EC 4.1.3.8) is a key enzyme in sugar metabolism and fatty acid biosynthesis. Its substrates and products are key intermediates in sugar metabolism and can serve as substrates for fatty acid synthesis. Additionally, ACL plays a significant role in plant growth and development as well as in enhancing plant stress resistance. Moreover, ACL is a key enzyme in the tricarboxylic acid cycle. ACL catalyzes the cleavage of citrate into acetyl-CoA, oxaloacetate, adenosine diphosphate, and phosphate in the presence of ATP and Coenzyme A. Malate dehydrogenase further catalyzes the conversion of oxaloacetate and NADH into malate and NAD+, and the rate of NADH decrease at 340 nm can be measured to determine the ACL enzyme activity.
