Aspartate aminotransferase, commonly known as glutamate oxaloacetate transaminase, abbreviated as AST or GOT (EC 2.6.1.1), is widely present in animals, plants, microorganisms, and cultured cells. It catalyzes reversible transamination reactions and is an important enzyme in amino acid metabolism. GOT/AST catalyzes the transamination reaction between aspartate and α-ketoglutarate, producing glutamate and oxaloacetate, which further decarboxylates to pyruvate; pyruvate reacts with 2,4-dinitrophenylhydrazine to form 2,4-dinitrophenylhydrazone, showing a brown-red color in alkaline solution. The activity of the enzyme can be calculated by reading the absorbance at 520nm.
