Asparagine synthetase (AS, EC 6.3.5.4) is widely present in plants and microorganisms and is a key enzyme that catalyzes the synthesis of asparagine from ammonia or glutamine and aspartic acid. This enzyme is of great significance for studying plant nutrition. AS catalyzes the transfer of the amino group from glutamine to aspartic acid to form asparagine, while producing glutamate. Utilizing the action of glutamate dehydrogenase on the generated glutamate, NAD+ is reduced to NADH. By measuring the increase rate of NADH at 340nm, the activity of asparagine synthetase can be determined. The enzyme-catalyzed reaction is: ATP + L-aspartate + L-glutamine + H2O = AMP + diphosphate + L-asparagine + L-glutamate.
